Solid-Phase Assembly Methods

Three approaches for assembling DNA fragments encoding TALE repeat arrays on solid-phase surfaces have been described in the literature (Reyon & Tsai et al., Nat Biotechnol. 2012; Briggs et al., Nucleic Acids Res. 2012; and Wang et al., Angew Chem Int Ed Engl. 2012). Because these methods are performed on a solid-phase, they enable assembly to be carried out using multi-channel pipets or automated liquid handling robots.
